Test Principle

Urea is enzymatically hydrolysed by urease:

Urea + H₂O → Ammonium ions + CO₂

The resulting ammonium ions react with α-ketoglutarate and NADH in the presence of GLDH.

During this reaction, NADH is oxidised to NAD+, leading to a decrease in absorbance at 340 nm.

The magnitude of the absorbance change is proportional to the concentration of urea in the sample.
